When creating a new Eyefinity configuration under Windows 10 64-bit with the 16.5.2.1 drivers I am seeing a problem. DisplayFusion does not recognize the Eyefinity configuration and auto-split doesn't do anything.

Could you attach a copy of your troubleshooting info when it's broken? Here are the steps:
  • Open the Settings > Troubleshooting tab
  • Click the "Copy to Clipboard" button
  • Paste the text into a text file (please don't paste the text directly into your reply, the formatting gets garbled and makes it difficult to parse)
  • Reply with the file attached

I'm having another issue with Eyefinity (but unrelated to DisplayFusion). After a reboot the Eyefinity displays are no longer present in Windows. In fact the AMD drivers don't even recognize the card properly. I have to do a DDU uninstall from Safe Mode and re-install to restore normal function. I've contacted AMD about this.

I suspect DisplayFusion isn't at fault!

Yeah, DisplayFusion shouldn't be able to affect the display drivers at all. You can test by disabling DisplayFusion from startup to see if that makes any difference :)
