Unfortunately, full screen game window lock the mouse cursor to the screen, and DisplayFusion can't help release it.
However, if the game has a "Windowed Mode" option in the video settings, you should be able to maximize the game window and still be able to move the mouse outside of the game window.

Actual Multiple Monitors has this feature, are you going to implement it one day?
We definitely won't be able to for the next release, but we'll add it to our list and investigate it for the release after that. I'll be sure to post an update when we have one.

Just wanted to follow-up and let you know that this feature is now available in the latest DisplayFusion Beta version (http://www.displayfusion.com/Download/Beta/).
There's now a built-in Function called "Prevent Window Deactivation." Just assign a key combination to it, and then run that key combo once you're in the game, and the game should stay active, even when working with windows on another monitor.

Tried to do as described, did not seem to work for me, set command to Ctrl + Win + Z
Played several games, tried to click on 2nd monitor, it minimized every game still. Cool feature though! Can't wait to see it in action.
Ctrl + Win + Z is actually one of the built-in key combinations (for the "Move Window to Next Monitor and Size Proportionally" Function). It shouldn't let you assign the same combo to two different Functions, but could you double-check the DisplayFusion Functions tab to make sure that Ctrl + Win + Z is only assigned to one Function?

I moved "Move Window to Next Monitor and Size Proportionally" to Ctrl + Alt + Right so Ctrl + Win + Z is not double bound.

Testing a few for you right now,
Splinter Cell: Conviction
some Source Engine games:
Team Fortress 2
Portal 2
Saints Row: The Third
The Elder Scrolls V: Skyrim
Assassin's Creed: Revelations
So I'm going to go into game play, press the key combination and try to move the mouse to my other monitor. If that doesn't work I will hold the key combination and try again. If still no luck, I will pause the game and try both again. I will let you know in a few moments
Yeah it should just work when you enter the game and press the key combo. However, I've just tested Saints Row 3 and it's definitely not working there. We're going to do some more investigating to see what we can find out.

I know this doesn't help in the functionality of DF, but wanted to let the OP know what I do for this situation.
Most recent games have a setting for Fullscreen Windowed Mode. This will allow you to go back and forth between monitors.

Team Fortress 2 worked from the pause screen, all the others I had no luck in. It does seem to be doing something as if I press while focused on a normal window like a web browser, it locks to always on top.
TF2 allows you to mouse off the game in the pause screen at any time, but if you click off, it would normally minimize. After pressing the key combination, it does not minimize if I click on another window.
It seems to work for keeping focus, but it is not releasing the mouse from the active game window.
