I made the plunge to a 43" 4k display and have been using display splitting to manage the display.

As I've used it, I've found that the top 2 quadrants are harder to use for regular work because they're kind of at too much angle for comfort. I've dealt with this by expanding the bottom two quadrants vertically to 1920x1200 and using the top quadrants as status/background displays.

What I'm finding myself doing a lot as I get accustomed to this size display is manually sizing some windows to take up about 75% or so of the screen when I have to work on something intensively and want a lot of screen real estate.

It would be great if I could define this as a "split" so that I could easily zoom windows to this region. It's not entirely logical from a pure splitting perspective, because I still want my 1920x1200 bottom quadrants and my 1920x960 top quadrants, and I don't think I can define a region with the splitting dialog as it is.

It's not possible to add an overlay split like that, but you could create a Custom Function to do what you're looking for:
  • Open a window and size it to the 75% size you want
  • On the Settings > Functions tab, click Add Custom
  • Drag the orange target icon and drop it on the window from step 1
  • Give the function a name and optionally a hotkey and click OK
  • Optionally assign it as a TitleBar Button by selecting it in the list and choosing "Toggle TitleBar Button"
  • Click OK to save the Settings
