I have triggers moving some application windows automatically on launch or activation. All was working perfectly till version 11.1b8.

In "Window created", WhatsApp and Discord, and in "Window focused", all of them.

If you disable the "Monitor Profile" match condition in those triggers, does it start working?

Yep, disabling the monitor profile specification did help to reactivate the automation.
Could it be because I altered my secondary monitor position in Windows settings?
It could be, if you open the Monitor Configuration window, does it show the profile is auto detected in the upper left corner? If not, it won't see the match condition for the trigger.
Re-saving your monitor profile should fix that up though.

I had to make the same position modification in the profiles that I did in Windows settings for my secondary monitor then save them. So I suppose that the saved profiles aren't linked to Windows settings and have to be manually modified one by one or did I miss something?
I'm not too sure what you mean, DisplayFusion will check the monitor orientation to see if a profile matches. If it's been adjusted after you saved the profile, each profile will need to be re-saved with the correct position
